I have always wanted to do this and am seriously contemplating it for next Summer. I'm on the East coast so would be heading somewhere on this side of the US. Anybody have any input/ recommendations?

Go to the doctor before you go and get a script for motion sickness. You do not want to get sea sick.... trust me. I've lived and played on the water most of my life but the one time I did get sick I wanted to jump overboard and swim to the bottom. It was ugly.

Its a blast if you are put on fish. Make sure you check out the boat and captain. Go to the docks where you plan to charter and see whats what before hand.
 
Deep sea fishing is great but if your not sure if you will get sea sick or not get the prescription for motion sickness. I have been on a few charter trips where people got sick and they were miserable.

If you are planning to stay on the east coast there are plenty of charters from half day trips to over night trips, and for all types of fishing. Search the internet for charters in the area you want to fish. ONce you find a couple of charters you are interested in ask plenty of question about the type of fishing you are looking for and when is the best times. Differnet fish have different peak seasons.

I fished a couple of charter trip about 10 years ago out of Florida and they were great fun. Good Luck!

Pricing is all across the board.... depends on what you want to do. I can't speak for the northeast coast but the southeast and/or the gulf coast you can go deep sea fishing for as little as $50-$75 on some of the party boats. Of course you will be standing elbow to elbow with 25+ complete strangers and are assured of a few tangled lines but you get what you pay for. Private charters depend on the boat and type of fishing you want to do. Pricing will start around $500 and go up from there. Last time me and a few buddies went we chartered a 42 ft hatteras for $850. Split 5 ways it wasn't bad....especially when you win the big fish pot. :)

Also, Jonesy has the right idea on going south and making a trip of it. The further north you go, the longer of a ride it is out to the Gulf Stream where the fishing is. Which means less time fishing if you have to work for your limit that day.

Doghouse Sportfishing ( link) is one of the better ones I have been on. He's located in the Outer Banks area.

The later in the fall/winter is when you want to go for Tuna and King Mackerel. Summer is when you can catch some Tuna and Dolphin.
 
I have gone twice while in Ft. Lauderdale and I had an absolute blast! I usually go with my lady and a couple uncles that live down there, so its usually around $500-$600 split three ways. Sure can get expensive, but really I dont think you can beat a day out at sea fishing for the big one! I recommend you at least try it out...sure it can be expensive but to me it's certainly worth the price. Oh and find out before you go if you have to also purchase the bait...some places will go out and spend time catching bait or you can pay for it from the local guys and just get right down to fishin'
 
Definitely the Florida Keys. With the saturation of charter boats down here, prices are very competitive too. Although, admittedly, I don't have to charter, but I know of some captains who run a few. Plus you have the added benefits of being able to go deep sea and hitting the backwaters as well (depending on the size of the boat of course - will need a smaller boat to get back into the bay waters). Getting into the time of year where the Wahoo are starting to run as well, and that's a hell of a fun fish to line.
